A noncompetitive inhibitor binds an enzyme away from its active site, at an allosteric site, changing its shape so the active site binds substrate less well.

Excess ATP binds an early glycolytic enzyme away from its active site, distorting its shape and slowing the pathway down.
Watch for
Because it binds elsewhere, a noncompetitive inhibitor's effect does not weaken as substrate concentration rises.
